Often cited as one of the best sci-fi movies of all time, 1982's Blade Runner was originally conceived to launch a series surrounding the Philip K. Dick novel Do Android Dream of Electric Sheep? The other films never came to fruition until now.
No plot, character, or casting developments have been revealed of yet, but Scott has stated that the new film will indeed be a Blade Runner sequel, and not a prequel.
Scott is currently awaiting the release of his highly-anticipated Prometheus, which originated as something of a prequel to Alien (watch a clip introducing Prometheus' David for a preview of the film). Might this Blade Runner sequel eventually diverge into its own film as well?
